New Protos 3 Colony Counter

New Protos 3 colony counter and rapid ID system saves time and provides accurate, traceable results in microbiology

Don Whitley Scientific Limited introduces a vibrant new automated colony counting and chromogenic identification system, Protos 3. This system allows accurate and reproducible identification of colonies cultured on chromogenic plates.

What makes the new Protos 3 outperform other commercial colony counters is the system’s ability to count colonies in seconds and identify microbial species by their colour on chromogenic plates. This is a great time saver, providing accurate, objective and fully traceable GLP compliant results.

The stylish, yet practical Protos 3, which comes in bright red, attaches easily to a computer and requires minimal training to set up. Users simply input their plate identification and click. The Protos 3, featuring a highly sensitive CCD camera combined with unique three colour LED lighting, rapidly images an infinite number of colony colours on one plate and detects colonies as small as 0.043mm.

The Protos 3’s powerful software then generates true to life counts and plate images, which can be transferred and stored in Excel. This GLP compliant process, with its full audit trail eliminates keying and image transfer errors providing accurate, objective data, which can be reviewed anywhere and anytime.

In this video – the latest in our series of customer testimonials – Dr Ayse Latif, Faculty of Medical and Human Sciences, University of Manchester, discusses her current research of exploiting tumour cell energy metabolism in order to improve gynaecological cancer treatments.

Dr Latif uses both a H35 HEPA Hypoxystation and an i2 Instrument Workstation. Whilst cells are initially incubated in the H35, they are then moved into the i2 via the Whitley Transfer Tunnel without exposure to ambient conditions, where they can be studied with a Seahorse XF Analyzer.

The i2 was designed to meet the exact requirements  of Seahorse Bioscience, meaning that the combination of this workstation and a Seahorse XF Analyzer permits simultaneous, real-time analysis of mitochondrial respiration and glycolysis in mammalian cells under precisely controlled conditions.

Huge Capacity with the A95TG

The Whitley A95TG (three gas) Anaerobic Workstation is currently the largest model in our range, with a maximum capacity of between 1000 and 1400 x 90mm Petri dishes. It is also extremely cost-effective; with an operating cost saving of 30% compared to the dual gas option.

The large, integral 30 litre airlock with automatic internal door is 20% faster than our previous MG1000 model, with the cycle taking just 5 minutes.

Each porthole also acts as a mini-airlock so you can transfer 10 plates through each time you enter or exit the workstation, thereby reducing unnecessary airlock cycles when processing small sample quantities.

H35 HEPA Testimonial Video

In this video, Dr Michael Cross from the MRC Centre for Drug Safety Science, University of Liverpool, discusses his current research involving the incubation of cardiac spheroids.

Dr Cross uses a Whitley H35 HEPA Hypoxystation, and explains the benefit of the HEPA filtration system in providing a sterile environment for the prolonged culture of cells in physiologically relevant conditions.

Hypoxia and Cancer Metabolism

An ever-increasing number of scientific papers are being produced that highlight the effects and importance of hypoxia in various research fields. Dr Burga Kalz Fuller, Product Manager at our American distributor, HypOxygen, has summarised five interesting and recent papers concerning hypoxia and its role in cancer metabolism research:

“Hypoxia signaling pathways in cancer metabolism: the importance of co-selecting interconnected physiological pathways” Masson & Ratcliffe, 2014.

In this paper, the role of hypoxia – as mediated by HIF, interfacing with many different oncogenic pathways – is reviewed. Activated HIF both suppresses mitochondrial metabolism and up-regulates biosynthesis of intermediates needed by proliferating cells in cancer. Complex relationships between HIF activation and cancer phenotype are discussed in detail by these Oxford UK researchers.

Cell Memory of Hypoxia in Metastases

This paper, entitled “Cellular memory of hypoxia elicits neuroblastoma metastasis and enables invasion by non-aggressive neighbouring cells”, was recently published in Oncology, and investigates the link between the level and duration of cells’ exposure to hypoxia and their ability to form metastases.

The researchers acknowledge that oxygen tension is not homogeneous throughout a tumour (as there will be areas of higher and lower oxygen tension within it) and thus seek to investigate the metastatic potential of cells that have been cultured hypoxically in vitro in comparison to those that have not, when implanted into a chick-embryo model.

Their results showed that cells with hypoxic preconditioning formed metastases in vivo, but cells without hypoxic preconditioning did not. These results support the discovery that the cells which were exposed to hypoxic preconditioning have a “memory” of this previous environment when introduced to normoxic conditions.
